Mechanism of Action: The medication works by blocking the PARP enzyme, which is crucial for repairing damaged DNA in cells. By inhibiting this repair mechanism, Olpaza prevents cancer cells from mending themselves, leading to their death and slowing tumor growth, especially in cancers with specific genetic mutations like BRCA.
Uses: It is used as a monotherapy or in combination with other anti-cancer medications, typically as a maintenance treatment after a patient has responded to platinum-based chemotherapy for:
Ovarian cancer
Breast cancer (HER2-negative, BRCA-mutated)
Pancreatic cancer (BRCA-mutated, metastatic)
Metastatic castration-resistant prostate cancer (with specific HRR/BRCA gene mutations)
Administration & Important Precautions
Dosage: The typical dosage is two 150mg tablets (300mg total) taken twice daily (approximately 12 hours apart), with or without food.
Administration: Tablets should be swallowed whole; do not crush, chew, dissolve, or split them.
Monitoring: Regular blood tests are necessary during treatment to monitor blood cell counts (red blood cells, white blood cells, and platelets) and liver function, as the medicine can lower these counts.
Dietary Restrictions: Avoid consuming grapefruit or grapefruit juice, and Seville oranges, as they can increase the levels of the medicine in the blood and raise the risk of side effects.
Pregnancy/Breastfeeding: Olpaza is not recommended during pregnancy or breastfeeding due to potential harm to the baby. Women of childbearing potential and men with female partners of reproductive potential must use effective contraception during treatment and for a period after the last dose.
